This ability has been, so far, used mostly for genome editing. Researchers at the Massachusetts Institute of Technology, however, have developed a unique use for these designable DNA-binding proteins programming them to detectspecific DNA sequences, such as pathogen-related DNA, which then activates a customized cellular response, such as cell death. The research team, led by James Collins, a professor of medical engineering and science at MITs Department of Biological Engineering and Institute of Medical Engineering and Science, and postdoctoral associate Shimyn Slomovic, published their results in a recent issue ofNature Methods. One might compare it to an alobere extremitt ordnungsprinzip that is equipped to contact security for a smart response when an intruder is detected, states Slomovic. How It Works This cellular detection ordnungsprinzip uses a type of DNA-binding protein known as zinc fingers, which can be programmed to recognize any DNA sequence. The team then built in an action response that is triggered by the recognition event. At left, cells glow red to indicate that the detection system has been successfully delivered. The system was designed to produce green fluorescence in cells carrying a viral DNA sequence, as seen at right. Image Shimyn Slomovic Synthetic zinc fingers derived from mammalian proteins were combined with intein proteins derived from yeast (inteins are short proteins that can be inserted into larger proteins, splitting them in half). The split protein pieces only become functional after the intein removes itself, allowing the two parts to join together and form a functional protein, which then activates a designated gene. A fluorescent protein (GFP) der ived from jellyfish was linked to the zinc fingers recognition of a DNA sequence from an adenoviral cell infected with this virus would then glow green. At the onset of the study, we envisioned a platform that was sufficiently robust to operate in a human cell, yet also specific enough to distinguish between a pathogen DNA target and the normal genome present in the cell, says Slomovic. It was exciting to reach a point in which the system could positively identify and report the infection of live replicating adenovirus inside living cells. Infected cells could also be killed. The system was programmed to produce proteins that alert immune cells to fight the infection (instead of simply identifying them by using GFP). Detection of the DNA target was linked to increased production of an enzyme called NTR. This enzyme activates a harmless drug precursor called CB 1954, which the researchers added to the petri dish where the cells were growing. When activated by NTR, CB 1954 kills the c ells. Since this is modular, you can potentially evoke any response that you want, says Slomovic. The cell can be programmed to kill itself, or to secrete proteins that will allow the immune system to identify it as an enemy cell so the immune system can take care of it. Future Applications Collins and Slomovic are now adapting their technology to detect latent HIV proviruses, which remain dormant in some infected cells even after treatment. Latent HIV provirus is pretty much the final barrier to curing AIDS, which currently is incurable simply because the provirus sequence is there, dormant, and there arent any ways to eradicate it, Slomovic says. Another possible use of this system would be assisting in the production of cell lines that scientists have engineered to contain critical genomic edits, such as resistance to HIV infection. Positive cells could be rapidly identified, streamlining this process. We also think the platform could be used to detect RNA-based viruses or used f or diagnostics in patient samples. In the near-term, however, perhaps the most immediate use of the system will be as a diagnostic tool that allows researchers to verify the results of experiments that are attempting to genetically alter types of cellular material. Yet another application would be keeping bioreactors free from pathogens. Many important therapeutic drugs and vaccines are generated in large bioreactors in which mammalian cells are cultured to produce large quantities, adds Slomovic. Bioreactors are susceptible to pathogen infection and a DNA sense-and-respond system could help arm these valuable cells with ways to prevent or contain such infections. Ward also talked to maternity leave expert Lauren Smith Brody and began internal conversations with people on her leadership team about the policy.Through her efforts working together with leadership and HR, they were able to increase the maternity leave policy from four weeks paid and eight weeks unpaid to before Ward went out on leave to have her daughter six weeks paid, six weeks unpaid, which I was able to take advantage of.A second revamp, all-inclusiveWhen Ward returned to work, she knew she wasnt finished overhauling the maternity leave. I almost immediately started working on revamping the policy again, she says, because in those conversations, one of the things I was told is, Its not necessarily in the budget for us to do it now, but lets keep talking about this and see if we can revamp it again in the future.So she kept talking about a second overhaul. I reignited that conversation early last year and, by the middle of the year had approved a new 12-week fully paid policy. And one thing that Im particularly proud of is that its not necessarily particular to men or women, or primary caregiver versus secondary caregiver, which I know is something a lot of companies do. Its just a blanket family leave policy. It was very important to me that it was LGBTQ friendly, so it was adoptive parent-friendly. And it didnt necessarily just favor birth mothers.During the second overhaul, Ward also put together a working group of parents and non-parents from different levels and d isciplines across the agency to figure out what types of resources were needed for manages, the partie taking leave, and their team.The bigger piece is making sure our employees are supported throughout the planning process, leave, and transition back to work, she said.Not just an office momWard felt the difference in going from an employee to a working mom and she worked to make sure she was taken just as seriously after she had her daughter as before.Its very hard to find your voice in the office and make sure that youre taken seriously as an employee once you become a parent, she says. One of the things that I am constantly telling people is that, in my own opinion, working has made me a better mom because Im more patient. I appreciate the time I spend with my daughter. When Im not with her, shes in school, shes learning, and shes growing and spending time with other kids her age.Working parents, she said, sometimes vilified as distracted and prone to needing time off, are actua lly excellent hires.Personally, I think that you can not make a better hire than a working mom, Ward says. Balancing Bot Leads the Way Balancing Bot Leads the Way Balancing Bot Leads the WayIt all started 12 years ago with the ballbot, a robot with a tall, thin body that moves in any direction, responds to a slight push, and balances atop a sphere slightly smaller than a bowling ball.Though the robot is perfectly suited to helping the elderly or disabled within their homes, researchers knew the rolling mechanism could be simplified to reduce maintenance and cut costs.Enter SIMbot. Researchers at Carnegie Mellon University recently improved upon their robot.Ballbot inventor Ralph Hollis calls the ball-balancing design a new, fundamental method of locomotion for robots. Hollis and Masaaki Kumagai, a prof of engineering at Tohoku Gakuin University in Japan, designed SIMbot along with graduate students.Yet that ball-driving mechanism is rather complicated, says Hollis, a professor at CMUs Robotics Institute. It uses five motors and there are belts and friction elements. So thats where SIMbot comes in.Though SIMbot relies on a rolling mechanism similar to ballbot, it includes just one moving part the ball.SIMbots spherical induction motor (thus the SIM) eliminates the mechanical drive systems used on previous ballbots. The new motor can also move the ball in any direction using only electronic controls. Image Carnegie Mellon UniversityBalance Is KeyWhat weve been trying to do with ballbot for a long time is to show it can do anything any other mobile robot can do, but also do a lot more because of the balancing and the omnidirectional behavior, Hollis says.Weve shown it can roll at about two meters per second faster than a rolles fast walk. And we can put arms on it so it can lead a person through a room, navigating while applying urging forces to the person while balancing, he adds.Balance is key, Hollis says. Sho uld the person the robot leads stumble or change direction, the robot remains balanced and upright. That means it can lead and guide disabled people, such as those who are blind or have difficulty with mobility. The ballbot can also lean over to assist a person in rising from a chair, carry packages, place items on a tabletop, and carry items on a tray-like surface among other tasks.But unlike ballbots complicated drive system, SIMbot has almost no mechanics. Basically, SIMbot is a robot made up of a body and a ball, Hollis says. The frame is a copy of ballbot and we demonstrated that it could have performance similar to ballbot.Whats more, because of its mechanical simplicity, SIMbot requires less routine maintenance and is less likely to suffer mechanical failures says Michael Shomin, a Ph.D. student in robotics who is part of Holliss team.The new motor can move the ball in any direction using only electronic controls. The rotor of the spherical induction motor is a machined, holl ow iron ball with a copper shell. Current is induced in the ball with six laminated steel stators, each with three-phase wire windings.Belts that drive the rollers wear out and need to be replaced, he says. And when the belts are replaced, the system needs to be recalibrated. The new motors solid-state system eliminates that type of maintenance.The six stators generate travelling magnetic waves in the ball, causing the ball to move in the direction of the wave. The direction of the magnetic waves can be steered by altering the currents in the stators.Future RedesignThough SIMbot does move as fast as the ballbot, it uses twice as much power to function as does its mechanical counterpart.Hollis and Kumagai intend to redesign the ball using different materials and making other design and software changes to improve motor efficiency. They also expect costs for the mechanically simple motor to sache in comparison with the ballbot motor.The tall and skinny SIMbot can be just as helpful to people as Hollis expects the ballbot to be. Really, these types of robots are ideal human helpers, he says.They cant get trapped in any cluttered space, they can go through narrow doorways. And if they run into something they can yield, he says. And with arms, they assist people mechanically. It saps your confidence. It can strain your relationships not to mention crush your productivity and morale. Recently I had the chance to weigh in on this critical issue with Juliana Reyes ofthe Philadelphia Inquirer, in a piece where 6 brave women shared their pay inequity stories. Try asking how much theyre hoping to land in terms of a percentage of their raise or how much they made when they first started at the job. You can also ask the range the y hope to fall within at review time. It helps if you have evidence to share,such as the latest salary report for your industry. Remember Employers cant legally tell you not to discuss salary. While youre at it, check out industry data from trade associations, industry competitors, and websitessuch asGlassdoorandPayscale. If a headhunter calls you with an opportunity (even if youre not interested), always talk to the person, as its a great way to see what employers are offering for someone *like you* right now.Prep before you makeyouraskSchedule a meeting with your manager just as you would for an important project. Come in prepared and ready to make an objective case for yourself. Data is important here. But so is framing. Dont open with talk about the wage gap or what your male colleague fruchtfleisch is making. (If you do, it could encourage your manager to begin telling you all the reasons Mark is wonderful or exceptional). Instead, focus on yourself. Have a list of your accompl ishments at the ready. But make sure to give yourself time to prep. Even if youre rightfully enraged about being underpaid, a drive-by negotiation rarely ends up getting anyone the results they want.Figure out a plan in case you get a noIf you get pushback, dont slink away defeated. Engage your boss and elongate the conversation. One woman executive I talked to described getting a no, and then asking what, exactly, her boss needed that he wasnt getting in terms of her skills. Pushing for that specific information, and then working on it, landed her a major promotion and pay raise not long thereafter. Another way to frame this is, What are the conditions that need to be true for me to move up/increase my pay?Analyze your wage gap with a lawyerIf you feel youre being discriminated against, dont be shy about talking to an employment lawyer. Do your own research and study theEqual Employment Opportunity Commissionswebsite(itsthe agency that enforces the Equal Pay Act). Familiarize yours elf with the Commissions rules For example, if you want to allegediscrimination, you have to compare yourself to another role thatsconsidered substantially equal. Know that if you formally file a complaint, your name will be attached to it. Its important to first read about others who have fought a wage gap and been through the process so that you can make an informed decision.Thankfully, not all companies are turning a blind-eye to pay inequity.
